At varying time intervals 100ml orange juice was
poured into sterile Stomacher bags and homogenised.
Samples (10ml) were collected and diluted 1:10 in a sterile
tryptone-salt solution. Yeast concentration was
determined on Yeast Glucose Chloramphenicol Agar
(serial dilutions were spread on a surface plate and incubated
at 30C for 72 h).
