From each fraction collected by HPLC, 50
µl of the methanolic suspension were dried under a stream of nitrogen and 0.5 M H2SO4 in
double distilled water (100 µl) was added and
vertexed. The solutions were incubated at 100ºC
for 1 hour. After incubation, double distilled water (1.0 ml) was added, passed through a SepPak Vac 1-cc C-18 cartridge (Waters). The water elute (800 µl) was collected and identified by
paper chromatography (Markham, 1982) using
different sugars as standards.
