2.3. Fruit sampling
At harvest and ateach removal, each fruit of the 10-fruit replicates
was cut to provide a 1 cm wide vertical slice. Two plugs (1 cmdiam.)
of tissue were taken from the stem and calyx ends of the slice and
frozen immediately in liquid N2 to provide 20 plugs from each tissue
zone for each replicate. Frozen tissues were cryogenically milled to a
fine powder using an analytical grinding mill containing liquid nitrogen, and stored at
-80 C untilused for analysis of enzyme activities and phenolic concentration.
