The analytical sensitivity of LAMP and nested-PCR assays was evaluated against 10-fold serial dilution of T. gondii tachyzoites DNA from 1 ng to 0.01 fg. The detection limits of the RE, B1-LAMP and RE, B1-nested PCR assays were one fg (Fig. 2A), 100 fg (Fig. 2B), 1 pg (Fig. 2C) and 10 pg (Fig. 2D) of the T. gondii DNA, respectively. The specificity of LAMP and nested-PCR assays was tested using the DNA samples of other parasites and human chromosomal DNA. No amplification was observed in the DNA samples of these groups (data not shown). Different DNA extraction methods did not affect the results of LAMP and nested-PCR.
