Indeed, the amount of acid soluble collagen present after the demineralisation of
powdered bone (,63 mm) for 2 h in HCl, was found to be three times the amount
generated from shards .2 mm in size [44]. Similarly, the apparently slower rate of
racemization of soluble proteins isolated from smaller sized bone particles [45], we
attribute to contamination with collagen due to pulverisation. From the results of the
studies described above, it is suggested that excessive grinding of samples should be
avoided to avoid damage to proteins and changes in amino acid profile of the soluble
fraction.
