2.5. Method validation
Method validation was performed under guidance from SANCO/
12495/2011 (SANCO, 2011) and the European Commission Decision
2002/657/EC (EC, 2002). The document is intended for official
control of laboratory monitoring of pesticide residues in foods, and
describes method validation and analytical quality control requirements
that validate data used for testing compliance with MRLs.
Five reagent blank samples and five blank control samples were
treated and instrumentally analysed to test a specificity of the
method. No target compounds were detected, which are corresponding
to the criterion of SANCO guideline (<30% of LOQ), and
proved its specificity. Analysis based on SRM transitions in this
study showed high selectivity, as demonstrated by signals below
30% of LOQ for each compound in blank samples. Accuracy, precision,
linearity, limit of detection (LOD), and limit of quantification
(LOQ) were studied as key parameters of method validation, and
the matrix effect was evaluated to accommodate unique characteristic
of ESI in LC–MS/MS analysis.
