2.3.7. Methylation of fucoidan
The deacetylated and desulfated fucoidan AmF3DADS wasmethylated using a modification of the previously described NaOHslurry method (Chiucanu & Kerek, 1984). In brief, a sample of thefucoidan AmF3DADS (2 mg) was solubilized in DMSO (1 mL), andpowdered NaOH (100 mg) was added to the solution, followed byMeI (0.2 mL). The mixture was stirred for 20 min, and NaOH andMeI were added again. This mixture was stirred periodically for1 h, and then it was cooled on ice. The reaction was terminatedby the addition of 1 mL of water. The excess MeI was removedby concentration under a vacuum, and the resulting solution waspassed through a Silica gel 100 C18 column (2.5 × 0.7 cm, Sigma,USA). Methylated fucoidan was eluted with 50% MeOH, concen-trated under a vacuum, and hydrolyzed with 2 M TFA at 100◦C for 6 h. TFA was neutralized with aqueous NH3(5%), and the result-ing solution was concentrated and lyophilized under a vacuum.Then, the resulting monosaccharides were reduced with NaBH4and acetylated with Ac2O in pyridine. Partially methylated alditolacetates were analyzed by GLC–MS.
