2.3.1. Determination of anti-inflammatory activity by
IL-6/luciferase assay on HeLa cells
HeLa cells were stably transfected with a luciferase reporter
gene controlled by the IL-6 promoter which is one of the target
genes for activated NF-_B. The cells were seeded into 24-well plates
and the extracts were tested in several concentrations (from 200 to
0.2_g/ml). As positive controls we used the solvents to dissolve the
samples (ethanol) and as negative controls we used unstimulated
cells. Parthenolide (Sigma) was used as a standard reference. The
enzymatic reaction was made with luciferase reagent (Promega),
measured and recorded using a Lucy-1 luminometer/photometer
(Anthos). More experimental details can be found in Bremner et al.
(2004, 2009).
