The previous record was just 140 patients.
The event was part of the Volunteer Doctors Project, jointly run by the King Prajadhipok's Institute (KPI), the Medical Council and the Public Health Ministry at Rajamangala University of Technology's Suvarnabhumi campus in Ayutthaya province.
KPI secretary-general Borwornsak Uwanno said it was the first time that so many volunteer doctors had participated in a mobile medical unit.
More than 300 volunteer doctors from 20 specialised fields provided treatment to 5,000 patients at the event.
But it was the 50 eye specialists who set the new world record.
Paisal Ruamviboonsuk, president of the Royal College of Ophthalmologists of Thailand, said many elderly people nationwide were suffering from eye-related diseases but lacked access to effective treatment.
The 50 doctors treated 450 patients within one hour, said Dr Paisal, himself a ophthalmologist.
Guinness World Records sent staff to witness the event and have certified the record
