AEE was carried out as follows: aqueous enzyme solution was
added to the kernel powder suspension (white pitaya kernels
was 6 g and water was 48 mL) to hydrolyze the pectin, cellulose
and protein in seeds. The amounts of enzymes used were: cellulose
– 50NCU g1dry matter, pectinase – 250PGU g1 dry matter, amylase
– 300U g1 dry matter, and acid protease –5AU g1 dry matter.
The suspension (pH 7.0) was incubated overnight (12 h) at the desired
temperature with constant stirring at 90 rpm. After enzymatic
treatment, half of the mixture was taken out and filtered.
The percolate was collected and extracted by hexane (20 mL) twice
in a pear-shaped separating funnel. The solvent was evaporated
and recycled.
