Minimum inhibitory concentrations (MICs) of plant extracts were estimated by well-established micro dilution broth susceptibility assay (NCCLS, 2002).
Testing was performed in duplicate. Stock solution was prepared by dissolving 100 mg of peel extract in 1 ml dimethylsulfoxide (DMSO) (100%) and was diluted to
obtain different concentration of extracts ranging from 10 to 100 mg/mL. 20 lL of the each dilution was added into the wells of 96-well micro titer plate containing 160 lL of Sabouraud Dextrose Broth (SDB). Then 20 lL of 5 105 CFU/mL (confirmed by viable count) of the fungal suspension was inoculated in into the each well. The plates were incubated for 24 h at 28 C and then shifted at 22 C to avoid overgrowth of the untreated controls. A micro titer plate reader was used to measure optical density at 620 nm and MIC was estimated in term of lg/mL.
