The presence of BaMV and satBaMV was
checked by dot or northern blot analysis with BaMV and
satBaMV-specific probes, respectively [19,35]. To ensure a fair
representation of the satBaMV population, leaves from several
individual bamboo plants in a given location were combined for
total RNA extraction (see below). Figure 1 shows the locations of
leaf samples and name designations for satBaMV populations.
To establish a baseline evolution in the greenhouse condition,
Nicotiana benthamiana leaves were co-infected with 0.1 mg
BaMV-S transcripts [36] and 0.1 mg BSF4 satBaMV RNA
transcripts [19] obtained by T7 RNA polymerase (Promega)
transcription, then harvested at 7 days post-infection (dpi).
