Fresh and young leaves of S. spontaneum were hand collected from individual selected plants as individual samples, and each sample was dried separately using self-indicating silica gel. These leaf tissues were taken back to the laboratory and stored in a freezer at 80°C for use. Genomic DNA was isolated from the collected leaf tissues using the modified CTAB method of Doyle (1991). Integrity and quality of DNA samples were evaluated by electrophoresis on 0.8% agarose gels containing 0.5 μg/mL ethidium bromide and quantified in a UV spectrophotometer.
