Culture Methods Practiced for S. costatum at the Tungkang Marine Laboratory Stock Culture
S. costatain is isolate and purified by using the micropipette method. The purified stock culture is then maintained in a 250 ml glass flask containing 100 ml of the F medium (Table 6.IV) but with no acration. The F medium is prepared in individual bottles by dissolving the proper amount of NaNO3 and NaH2PO4.2H2O,Na2SiO3 and micronutrients separately in distilled water after which they are autoclaved. One ml of each of these solutions is needed for 1 l of culture water. One hundred milliliters of autoclaved seawater and 0.1 ml of each of the three stock solutions are added to one 250 ml flask One ml of Skeletonema culture, which is not in decating is then inoculated. Mass Culture
One glass carboy containing 1 l of autoclaved seawater enriched with F medium is inoculated with one stock culture of 100 ml. At 25 to 30 C and 5000 to 9000 lux. Maximum growth occure after 3 to 4 day. One glass beaker containing 10 l of boiled seawater.enriched with F medium is inoculated with two to three stock cultures of 100 ml each,or,with 200 to 300 ml of 1 l culture. At 25 to 30 C and 2600 to 6000 lux. Maximum growth occurs after 3 to 4 days after which transfer is made to the large culture tanks. A fiberglass circular tank of one ton is filled sand filtered seawater to the 800l mark. Two 10-l quantities of Skeletonema culture,or 5 to 10 l of the concentrated algae collected from the Kaohsiung Harbor with a 150 mesh plankton net is then inoculated. The nutrient solutions are prepared by dissolving the proper respective amounts (Table 6,II,III)of KNO3 or CO(NH3)2 Na2HPO4.12H2O in 1 l of freshwater and Na2SiO3 in another 1 l of freshwater (if Na2SiO3 is dissolved in seawater , a white precipitate will form). These sub-stances are then all added the same morning to the culture tank before or after the inocula. After 2 to 3 days and brfore thedecay of culture beging,10 to 20 l of Skeletonema culture or 5 to 10 l of the concentrated Skeletonema collected with a 150 mesh net, is transferred to anoter one-ton tank and same procedure repeated as reported above. The one-ton cultures of Skeletonnema are used to inoculate 10-to 100 ton cultures by using a 01 to 02 ration of inoculation. The seawater of the large tank culture is prepared to contain a 25 to 30 ppt salinity by diluting with fresh water. Mild aeration by air stones is provided to the medium in order to avoid the breaking up of the diatom chiatom chains. The one ton tanks are covered urider a plastic roof , while the larger concrete tanke are outdoors. Shading or applying artificial light to obtain optimum light intensities is practiced.
