A combined culture, bacteriological water testing kit
(BWTK) and molecular tool (multiplex PCR) as water
testing kit has been designed. Water sample (15 ml) is
added in BWTK and kept at room temperature for 12 h to
observe for colour change. A non-potable sample is
depicted by colour change to yellow. Further, a PCR isperformed by taking an aliquot of 200 μl in a PCR tube,
heat treated at 95°C for 10 min. Further it is centrifuged
at 10,000 rpm for 5 min and 5 μl of supernatant used for
multiplex PCR.
