2.6.3.2. 1,1-Diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity. DPPH assay was done according to the method of Prior, Wu, and Schaich (2005) with some modifications. The stock solution was prepared by dissolving 49.29 mg DPPH with 25 mL methanol and then stored at −20 °C until needed. The working solution was obtained by mixing 950 μL stock solution with 25 mL methanol to obtain an 190 μM DPPH solution. 30 μL sample extracts or Trolox standard were allowed to react with 200 μL of the 190 μM DPPH solution and absorbance readings at 517 nm were taken after 10 min reaction using a multi-detection microplate reader (Biotek, Synergy HT, Winooski VT, USA) with Gen5 software. The standard curve was used between 37.5 and 600 μM Trolox and results are expressed in Trolox equivalent (TE; mmol/g dw).
