Single site-directed mutants were constructed following the
standard protocol using WT construct pET32a-VP as the template.
Easily oxidizable methionines Met152, Met 247, Met262, and
Met265 were replaced with high-redox-potential residues Leu
or Phe respectively. Ala77, Ala79, and Ile 81were replaced with
Ser/Glu, Ser/Leu, and Leu respectively according to the paper by
Miyazaki-Imamura et al. [24]. Pro141, Ser168, and Phe186 were
replaced with Ala respectively. Triple mutants A77S/A79L/I81L
and A77E/A79S/I81L were also constructed based on combinatorial
mutagenesis studies toward oxidative stability of MnP [24].
