For quantitative estimation of aflatoxins, measurements were
performed on HPLC-system in the Mycotoxin Laboratory, Department
of Food and Agriculture, Ghana Standards Authority, Ghana.
An HPLC apparatus containing Shimadzu LC software package
designed for HPLC real time and post-operative analysis operated
through computer, injection system, pump able to maintain a flow
rate of 1 ml/min, column heater able to maintain a constant temperature
of 40 C, chromatographic column c18, 4.6 mm 250 mm,
5ul or equivalent post-column derivatization system, and column
oven was used. Isocratic mobile phase consisting of water, methanol
and acetonitrile ratio (60:20:20) was used at a flow rate of
1 ml min1
. Injection volume 50 ml (sample injector with 50 ml
sample loop) was used. The eluent was detected using spectrofluorometer
detector set at emission 435 nm and excitation at 365 nm.
