Pharmacokinetic Analysis
A noncompartmental analysis was used to determine
the pharmacokinetic parameters using Drug and Statistics
Software version 2.1 (University of Science and
Technology, Hefei, People’s Republic of China). Individual
and mean plasma concentration–time curves
were constructed, and the Cmax and Tmax were obtained
directly via visual inspection of these curves. The AUC0–t
was calculated using the trapezoidal rule.20 AUC0–∞ was
calculated as follows:
AUC0–∞ = AUC0–t + Ct/ke
where Ct
was the last measured concentration at time
t, and ke was the terminal elimination rate constant
estimated by log-linear regression analysis on data visually
assessed to be a terminal log-linear phase. At least
3 points were used for estimation of ke. The apparent
terminal elimination t1/2 was calculated as follows:
t1/2 = 0.693/ke.
Interindividual variability for the considered pharmacokinetic
parameters was evaluated by %CV.
