For scale-up of SSF in a 2 dm3 flask with a working volume
of 1 dm3, yeast was added after 2 days PS saccharification
(Fig. 5a). The highest ethanol concentration was 11.34 kg m3
at 2 days after yeast addition, and initial glucose content in the
SSF was 23.79 kg m3, whereas the initial level of total
reducing sugars was 31.62 kg m3. Compared to the SSF
process in a 250 cm3 flask, the maximum ethanol concentration
was similar (Fig. 5b). Based on the saccharification yield of
73.5% and glucose ratio of 80% in reducing sugar from Fig. 3,
the reducing sugar and glucose concentrations from
50 kg m3 PS cellulose were estimated to be 36.78 and
29.0 kg m3, respectively. The ethanol yield was 80% (Fig. 3c),
therefore, 11.8 kg m3 ethanol would be expected, which was
reproduced on the 2 dm3 flask scale. The C8-8 cellulase
powder showed a high CMCase activity and was stable during
saccharification and SSF. The cellulase activity was
6.2 105 FPU m3 during saccharification, decreased to
5 105 FPU m3 because of yeast inoculum, and remained at
2.3 105 FPU m3 during SSF (Fig. 5a).
