It shows the combined effect of concentration of CMTKP (X1) and concentration of calcium chloride (X2) on particle size. It can be inferred that concentration of CMTKP is having more pronounced effect than calcium chloride concentration on particle size. Increasing the concentration of CMTKP was found to increase the particle size of the nanoparticles, which can be attributed to inadequate interaction of crosslinker and polymer and/or in increase in viscosity of the solution with increase in polymer concentration.Response surface plot showing combined effect of concentrations of CMTKP (X1) ...Fig. 2. At lower values of polymer concentration, increasing the crosslinker concentration resulted in increased entrapment of the drug which can be attributed to higher degree of crosslinking at higher crosslinker concentrations. Further, it can be observed that increasing the polymer concentration resulted in decrease in % encapsulation, which was more pronounced at higher crosslinker concentrations. In addition, at higher values of polymer concentration increasing the crosslinker concentration did not result in any significant change in % encapsulation. The optimization was done to locate the optimal concentrations of polymer and cross-linker under the constraints of minimizing the particle size and maximizing the % encapsulation. The numerical optimization tool provided us with different sets of optimal solutions. Using these solutions, an optimal batch of tropicamide-loaded CMTKP nanoparticulate formulation was prepared and analyzed for particle size and % encapsulation efficiency. The optimal calculated parameters were concentrations of CMTKP-0.10% (w/v) and calcium chloride-0.11% (w/v). The optimized batch of tropicamide-loaded CMTKP nanoparticulate formulation was found to have the particle size (Y1) of 339 nm (predicted 317.69 nm), and % encapsulation efficiency (Y2) of 15.57% (predicted-16.25%). The lower value of % prediction error (−1.09% for Y1 and 3.12% for Y2) indicates the reliability of developed mathematical models.The transmission electron micrograph (Fig. 4) of the optimized batch of nanoparticles reveals the ovoid morphology of the nanoparticles. The particle size and shape of the nanoparticle together determines the ocular irritation potential of the particles inserted into the cul-de-sac [25]. According to TEM micrograph size was determined to be 20–40 nm. Firstly, particle size analyzer is an assorted technique because of its low measuring range thus it is quite difficult to determine the presence of microparticulate drugs in nanosuspension whereas, TEM shows the particle as well as its size so it is easy to differentiate between microparticulate drug particles in nanosuspension [26]. Secondly, particle size analyzer measures the hydrodynamic diameter which not only depends on the particle core but also on surface structure, concentration and type of ions in the medium while, in TEM the particle is removed from its native environment [27].TSP has earlier been reported to increase the pre-corneal retention and intraocular penetration of ketotifen, diclofenac [28], gentamicin and ofloxacin [29]. In the present study carboxymethyl derivative of TSP have been evaluated as nanoparticulate ocular delivery system for tropicamide. Tropicamide is a poorly water soluble, weakly basic drug having pKa of 5.2 [30]. Because of its poor solubility, its aqueous solutions are buffered to pH of 5.0–5.5 [31]. Due to the lower pH of the formulation it shows poor ocular tolerance and bioavailability. To improve its ocular availability formulations solubilize
