The optimum HS medium containing 0.5% glucose,
1.5% fructose and 4% YE yielded 6.7 g/l of cellulose in
shaking culture, which was approximately 3-fold higher
than the yield in normal HS medium. However, addition
of 4% YE to the culture media is economically unfeasible.
Therefore it is necessary to further investigate cheap
organic nitrogen sources for economic production of
cellulose. Matsuoka et al. (8) reported that CSL was found
to be the most suitable organic nitrogen source for cellulose
production by A. xylinum subsp. sucrofermentans
BPB2001 and lactate in CSL promoted cell growth and
cellulose production. They reported that 4.04 g/l of cellulose
was obtained from CSL medium containing 4%
fructose in flask culture. In this study, CSL media,
which contained 0.5% glucose, 1.5% fructose, 0.27%
Na2HP04, 0.115% citric acid and various amount of
CSL, were examined in shaking cultures. As expected,
cellulose production increased with increasing CSL concentration
and reached a maximum of 6.61 g/f in 6% CSL medium (Fig. 4). This yield is almost identical to
that obtained in 4% YE-HS medium. These data indicate
that CSL can be successfully substituted for both
YE and peptone in HS medium.
