Liquefaction was carried out at 100 cm3 Erlenmeyer flasks
containing 25cm30.5cm3 of different concentrations of dried
sweet sorghum(as described later), in orbitalshakers (Zhicheng
ZHWY-211C) at an agitation rate of 180 r min1 and incubation
temperature 50 C 1 C. Solutions of dried sweet sorghum
were prepared in distilled water.Commercialenzyme solutions
used during this research were Celluclast 1.5L and Novozym
188 (Novozymes A/S, Denmark) at a ratio of 5:1 volumes with
a filter paper activity of 83 FPU cm3, as measured by the filter
paper assay [14]. As it will be discussed in Results & Discussion
section, Celluclast 1.5L was added in the start up of liquefaction
step while Novozym 188 was added at the beginning of
the fermentation.
