This studydemonstrates,throughaGWASapproach,anasso-
ciation betweengeneticvariationmappingtothe CYP2B6 locus
and plasmalevelsofmultiplePCBswithinapopulation-based
cohort. Therelationshipappearstobecomplex,witheffects
mediated predominantlythroughPCB-99andPCB-118.
The presentstudyisthe first examination,inhumans,ofthe
relationshipbetweengenome-widegeneticvariationandplasma
levels ofabroadrangeofPCBs.Ourresultssupporttheviewthat
CYP2B6 is involvedinthemetabolismofatleastsomePCBsin
humans, aspreviouslydescribedinexperimentalstudies(Gährs
et al.,2013; Lehmler etal.,2010). Furthermore,CYP2B6hasalso
been reportedtometabolisearangeofenvironmentalchemicals,
including insecticidessuchasmalathion(Buratti etal.,2005) and
carbofuran (Usmani etal.,2004).
CYP2B6 has severalknownalleles(http://www.cypalleles.ki.se/
CYP2B6.htm), manyofwhichareassociatedwithserumlevelsofa
rangeofpharmaceuticaldrugs. CYP2B6 genotypinghasbeenap-
plied successfullyinHIV-infectedindividualstoreducethether-
apeutic doseofefavirenz,resultinginimprovementofcentral
nervoussystem-relatedsideeffects(Gatanaga etal.,2007). The
commonest alleleis CYP2B6n6 [Q172HandK262R],taggedby
rs3745274andrs2279343,respectively.However,thesetagging
SNPs werenotassociatedwithserumPCBlevelsinthepresent
study.
All PCBsfoundtobeassociatedwithgeneticvariationin
CYP2B6 inthisstudyarehighlybioaccumulative.Onecommon
waytocharacterizethePCBsisthroughtheirbindingtothearyl
hydrocarbon(Ah)-receptor.Becauseoftheirstructure,dioxin-like
PCBs areagonistsofAh-receptors,whilenon-dioxin-likePCBsare
not. PCB-99,PCB-138andPCB-153arenon-dioxinlike,whilePCB-
118andPCB-105aredioxin-like. McFarlandandClarke(1989)
suggestedaclassification basedontheabilitytoinduceCYPsina
barbiturate-likefashion,andPCB-99andPCB-153belongtothat
class. Wolffetal.laterfurthersubdividedthePCBsintoanti-
estrogenenicandphenobarbital-typeinducers,andPCB-99and
PCB-153belongtothatlatterclass(Wolffetal.,1997). Itshould,
however,bepointedoutthatnotallPCBsconsideredtobephe-
nobarbital-type inducersdemonstratedanassociationwithge-
netic variationinthe CYP2B6 geneinthepresentstudy.Within
this groupitwasmainlythosewithamoderatenumberof
chlorine atomsthatwereassociated,butnotthosewithalarge
number ofchlorineatoms,suchasPCB-180andPCB-194,havinga
longerhalf-life(Wimmerovaetal.,2011).
Conditional phenotypeanalysessuggestthatassociationsig-
nals withpollutantsatthe CYP2B6 locus aremediatedthrough
PCB-99 andPCB-118.Theonlycommondenominatorforthese
pollutants, beingrelatedtovariationinthe CYP2B6 gene, seemsto
be thelownumberofchlorinesubstitutions,possiblyreflecting a
shorterhalf-lifethanthemorelipophilicPCBswithalarger
number ofchlorineatoms.ThesubstitutionpatternofPCB-99and
PCB-118withchlorinesubstitutioninboththemeta-andthepara-
position preventsdegradationinbiota(Furukawaetal.,1978). The
same istrueforthehexachlorobiphenylsPCB-153and-138.The
largermolecules,hepta-todecacholorbiphenylcongenersand
dioxin-likehexachlorobiphenylcongenersshownocorrelationvs
variationinthe CYP2B6 gene.
5. Conclusions
The presentstudyisthe first ofitskindtorelategeneticvar-
iation toplasmaconcentrationsofPCBsbymeansofaGWASap-
proach.Thisapproachhastheadvantagethatitcoversthein-
formation onvariationinallhumangenes,andtherebynoprior
hypothesesonwhichgenesthatareofimportanceareneeded.The
environmentalimplicationofthecurrentresultsisthatthe
CYP2B6 isinvolvedinPCBmetabolisminhumansandthat
