Cordyceps militaris has beenusedasamedicinalstuffinoriental
medicine forthetreatmentofinflammationandcancercom-
plications. Someimmunomodulatoryandantitumoractivitiesof
Cordyceps militaris crude extractshavebeenknown(Park etal.,
2005; Leeetal.,2006;Kimetal.,2008;Hanetal.,2010). How-
ever, itsactivecompoundshavenotbeenfullyexploredfromthe
anticancer andanti-inflammatoryviewpoint.Inthisreport,we
investigated theinhibitoryeffectsofcompounds 1–10 isolated
from thefruitingbodiesof Cordyceps militaris, onmacrophage-
mediated inflammatorymediatorssuchasNOandcytokines,TNF-
and IL-12production,andhumancancercellproliferation.
Increased levelsofNOhavebeenobservedinvariousimmuno-
logical diseases,whichmaybecomeself-destructive(Libby, 2007).
Since macrophagesareoneofthemajorimmunecellsthatproduce
NO oninductionofiNOSgene,theregulatoryeffectofcompounds
1–10 on itsproductionwasevaluatedinperitonealmacrophages.
In thisstudy,significantreductionsinNOlevelswereobserved
in LPS/IFN- induced condition(Fig. 2A andB).Ourresultsare
in agreementwithpreviousreportthatcompound 8 inhibits the
enhancedNOproductioninLPS-stimulatedmacrophagesbydown-
regulation ofiNOS,COX-2expressionviathesuppressionofNF-B
activation, Aktandp38phosphorylation(Kim etal.,2006). Onthe
other hand,cytokinesproducedbymacrophagessuchasTNF-and
IL-12 areknowntoplayacentralroleininitiatingandperpetuat-
ing avarietyofinflammatoryandautoimmunedisordersincluding
rheumatoid arthritis(Brennan andMcInnes,2008). Tosustainan
inflammatory response,anelaborateinteractionbetweenboth
innate andadaptiveimmuneresponseisrequired(Libby, 2007).
It hasbeenreportedthatnaturalextractsorcompoundsshowed
inhibitory effectsontheseinflammatorycytokinesareknownto
reduce theseverityofinflammatorydiseases(Murakami, 2009).
In thisstudy,whentheprofileofcytokinesecretionbyactivated
macrophages wereanalyzeditwasrevealedthatcompounds 8,
followed by 3, 7, 9, and 10 significantly suppressedinflammatory
cytokines TNF- and IL-12(Fig. 2A andB).Thisisthefirstreport
on thesecompoundsfortheirinhibitionofcytokinesTNF- and IL-
12 inLPS/IFN- induced macrophages.Previouslyitwasreported
that hotwaterextractof Cordyceps militaris significantly induced
the interleukin(IL)-18expressionsthroughitspromoteractivation
in mousebrainandliverRAW264.7cells(Kim etal.,2008). Itis
known thatLPSinductionofIL-18wasdifferentdependingoncell
type. InKupffercellsandperitonealcells,IL-18didnotrespondto
LPS, butIL-18wasinducedbyLPSinRAWcellsandmousespleno-
cytes (Novick etal.,1999). Incontrasttopreviousdata,thisresult
indicated thatcompounds 8, followedby 3, 7, 9, and 10 potently
