Quadruplicated samples of 50 mg were dispersed in 6mL of 2M KOH and energically
shaken at room temperature for 30 min. 3 ml of 0.4M Sodium acetate buffer pH=4.75 and
60 ~1 of amyloglucosidase (Ref 102 857, Boehringer) were added to this suspension and
incubated for 45 min at 60°C in a controlled shaking water bath. Starch was measured as
glucose with Peridochrom Glucose GOD-PAP (Ref 676 543, Boehringer). Factor conversion
from glucose to starch was 0.9.
