The challenge experiment was carried out by the cohabitation method. The A.
salmonicida SN1 strain used was isolated at the Institute of Parasitology, A° bo AkademiUniversity, Department of Biology, Finland; during a natural outbreak of furunculosis in
rainbow trout, A. salmonicida was grown aerobically for 24 h in 10 ml tryptic soy broth
ŽTSB. at 208C. The cells were harvested by centrifugation at 2000) g and washed twice
with 0.9% saline. The bacterial suspension was diluted to a density of 1.8)107
CFUrml, and 0.1 ml of this suspension was injected intraperitonealy into cohabitation
fish. The fish were anaesthetised with Benzocaine Žethyl-4-aminobenzoate. before
injection. All cohabitants Ž15 for each group. were marked by clipping the adipose fin
after injection, and were replaced into the appropriate tank with experimental fish. Dead
fish were collected daily and the cause of death was confirmed by microbiological
samples taken from spleen and kidney on TSA plates; the plates were incubated
aerobically for up to 3 days at 208C.
