Because interference with a particular
cell death pathway often results in a shift to an alternative cell
death pathway, one needs to combine these knockdown approaches
with analysis of the cell death type observed after knockdown.
This is exemplified by the finding that knockdown of RIPK1
shifts TNF-induced necroptosis to apoptosis; an additional knockdown
of caspase-8 blocks apoptosis and reverts the cell death
pathway to necroptosis again, which is dependent on RIPK3 [61].
