To obtain a comprehensive insight into the mycoparasitic
potential of Trichoderma, the transcription of chitinaseencoding
genes (subgroup A, B and C) and genes encoding
N-acetylglucosaminidases was assessed by RT-PCR from
different stages of dual agar cultures of F. culmorum or F.
graminearum vs T. gamsii 6085 and 6317. It is common in
confrontation assays to overlay agar plates with cellophane
membranes to facilitate the harvesting of the mycelia.
However, we wanted to exclude the potential influence of
cellophane on chitinase gene expression and therefore
developed an optimized RNA extraction method from agar.
