and cultured on potato
dextrose agar (PDA) in Petri dishes for 6 days at 25 C. The conidia
suspension of T. longibrachiatum was prepared by flooding each
dish with 3 ml of sterilized distilled water, and the agar surface was
scraped gently with sterile glass rods and collected in 250 ml
beakers. Eighteen Petri dishes with T. longibrachiatum were used to
collect sufficient conidia for the experiment. The suspension of
54 ml was then mixed using a hand mixer to separate and disperse
the conidia. The density of conidia was assessed using a
haemocytometer. Suspension of different densities, 1.5 107
,
1.5 106
, 7.5 105
, 3.0 105 and 1.5 105 conidia ml1
, were
prepared and stored at 4 C.
