2.4. HPTLC-photodensitometry
10μL each of the different concentrations of standard
solutions were spotted (band width: 6 mm) in triplicate on
HPTLC plates precoated with 0.2 mm layer of Si-gel Si60F254
(E. Merck) using an automatic applicator Linomat-5.
Chromatograms were developed for 9.0 cm using n-butanol:
n-propanol: glacial acetic acid: water (30:10:10:10 v/v/v/v).
Chromatography was performed in a twin trough chamber
(Camag, Switzerland). After development the plates were
sprayed with anisaldehyde sulphuric acid reagent and the
spots were detected by heating the plate at 105–110 °C for
5min. The developed plates were then scanned at 600 nm so
as to record the peak areas. The calibration curve of
aloeverose was obtained by plotting peak area vs. concentration
of aloeverose applied.
