Abstract Inthisworkweanalyzedtheeffectsofthreeculturesystemsondevelopmentalabilityofbovineembryosinvitroproduced withsexedsperm,thesurvivaltovitrification(cryologicvitrificationmethod)ofsuchblastocysts,andtheirpregnancyratesafter embryotransfertorecipients,bothasfreshandaftervitrification/warming.Finally,wemeasuredtheaccuracyofthesorting protocolbyapolymerasechainreaction-basedmethodtovalidatetheembryosexatblastocyststages.Weconfirmedanindividual effectofthebullaswellasdevelopmentratesofembryosproducedwithsortedspermlowerthanembryoswithunsortedsperm, independentoftheculturesystemused.Thecryoresistancetovitrificationofembryosproducedwithsexedspermdidnotdiffer fromthatofconventionallyproducedembryos(re-expansionratesat24and48h:74.6%vs.75.5%,and64.5%vs.68.1%for embryosproducedwithconventionalandsortedsperm,respectively;hatchingratesat48h:63.55%vs.55.5%forembryos producedwithconventionalandsortedsperm,respectively).Finally,nosignificantdifferenceswerefoundinpregnancyratesafter theembryotransferoffreshandvitrified/warmedblastocysts(52.8%vs.42.0%,respectively;P0.05).Maleandfemaleembryos producedwithsortedspermshowedthesamequalityintermsofdevelopmentalability,cryoresistance,andpregnancyratesafter transfer.Ourculturesystem,coupledwiththevitrificationinfiberplugs,providesgoodqualitysex-knownembryoswhichsurvive vitrificationatsimilarratesthanembryosproducedwithconventionalunsortedsperm;alsoitproducesgoodpregnancyratesafter transferofsexedembryosbothfreshandaftervitrificationandwarming. ©2012ElsevierInc.Allrightsreserved.
