Phytate biodegradation Samples (wheat bran, rice bran and
feeds) were autoclaved for 20min at 120°C to inactivate any
phytase present in them. Ten grams of the autoclaved sample was
suspended in 100 ml of acetate buffer (0.2 M, pH 5.5). The suspension
was incubated with 1 ml of cell suspension (3500 U/ml ) at
37°C on a rotary shaker (110 rpm, TAITEC Co., Tokyo). The reaction
was stopped at various times during the reaction by adding
an equal volume of 10% trichloroacetic acid and then the amount
of liberated inorganic phosphate was measured.
The feeds were obtained from a farm of Hiroshima University.
Feed 1 was for sheep. Feed 2 and Feed 3 were for broilers.
