Fig. 4. Substrate specificity (a) and kinetic properties (b) of amylase were
investigated. (a) The substrate specificity of the amylase was measured in the
presence of starch, glycogen, a/b-cyclodexterin, pulullan and maltooligosaccharide.
The substrates were prepared in 100 mM sodium acetate buffer at pH 5.0. Starch
was used as a reference substrate to compare the activity of enzyme. (b)
Lineweaver–Burk plots in the presence of different concentration of soluble starch
were studied. The activity was assayed in 100 mM acetate sodium buffer pH 5.0 at
70 C using DNS.
