Other cytokinins such as KT (kinetin), 2-ip (2-isopentenyladenin) and zeatin were used at 0.2, 0.5 and 1.0 mg/L, but none of these treatments induced shoot buds either from cotyledon or from hypocotyl explants of the three varieties. Callus growth and shoot buds regeneration were observed from cotyledon and hypocotyl explants of all the three varieties cultured on MS medium fortified with various concentrations of NAA (α-naphthaleneacetic acid, 0.2-1.0mg/L) and BA (0.2-2.0mg/L). On MS +NAA (0.5 mg/L) + BA (0.2-2.0 mg/L) callus formation and differentiation of 2-4 shoot buds per explant was obtained in about 40% hypocotyl cultures of Th-10 black and NP-9 black, cotyledons were more responsive than hypocotyls. In the variety Partial hull black, although good callus growth occurred, bud differentiation was observed only in 10% of cotyledon and hypocotyl explants. In general, shoot buds developed on NAA + BA medium were very small and most of them failed to develop into normal plantlet.
