An important variable in MCTS is tumor size since it is correlated with cell function, as well as drug penetration and transport. In general, spheroids between 200 and 500 μm are sufficiently large to develop gradients of oxygen, nutrients, and catabolites. Above a critical size of 400–600 μm, spheroids develop a central secondary necrosis where the innermost cells die of apoptosis or necrosis.
