which provides important information for establishing a robust tissue
culture system for sorghum.
To efficiently regenerate sorghum in vitro cultures, it is imperative
to introduce a management strategy to minimize the effect of short
duration of callus regenerability. It is necessary to continuously initiate
fresh IEs in order to maintain a highly efficient tissue culture system.
Sorghum lines, such as Tx430 and SA281, can grow in a temperaturecontrolled
glasshouse throughout the year facilitating the harvest
of fresh IEs at any time. Therefore, vigorous embryogenic calli can be
generated consistently allowing the optimized tissue culture system to
be utilized for transformation irrespective of growing seasons.
