In this study, optimization of the surface culture conditions of C. militaris for cordycepin production was carried out. Most of the cordycepin synthesized by C. militaris existed in the culture medium. For the medium condition, the mixture of peptone and yeast extract in which the yeast extract ratio was greater than 75% was suitable, and the optimal carbon/nitrogen ratio (w/w) was 2/1 when glucose and the mixture of peptone and yeast extract (P/Y = 1/3) were used as the carbon and nitrogen sources, respectively. As for the medium depth, the lower medium depth leads to a higher productivity in the 1.8–5.3 cm medium depth range, because the utilization efficiency of the biofilm decreases with an increase in the medium depth over this experimental range. The maximum concentration of cordycepin obtained in this study was 640 mg/l, which was considerably high compared to previous reports.
