HEK293T cells (8 x 105 cells/well)

HEK293T cells (8 x 105 cells/well) were seeded in a 6-well tissue culture plate. After
24 hours of incubation at 37C, the cells were co-transfected with plasmids pcHXB3Env and
pcDNA3.1Tat101FLAG using Lipofectamine®2000 according to the manufacturer’s instructions.
The transfected cells, termed HEK293TEnv, were further incubated at 37C in 5% CO2
for 24 hours. HeLa T4+ cells (8 x 105 cells/well) were seeded in a 6-well tissue culture plate
and incubated for 24 hours. Then, the cells were transfected with pTruLTRGFP using
Lipofectamine®2000 according to the manufacturer’s instructions. The transfected cells,
termed HeLaT4Tru, were further incubated at 37C in 5% CO2 for 24 hours.
