Substrate specificity of lipase tow

Substrate specificity of lipase towards various p-nitrophenyl
esters and triglycerides was examined at various temperatures
(37, 45 and 55 1C). The lipase activity towards
p-nitrophenyl esters was determined using the substrates
p-nitrophenyl butyrate (C4 : 0), p-nitrophenyl caproate
(C6 : 0), p-nitrophenyl caprylate (C8 : 0), p-nitrophenyl caprate
(C10 : 0), p-nitrophenyl laurate (C12 : 0), p-nitrophenyl
myristate (C14 : 0), p-nitrophenyl palmitate (C16 : 0) and
p-nitrophenyl stearate (C18 : 0) at a final concentration of
2.5 mM. The enzyme activities were assayed as described by
Krieger et al. (1999). Lypolytic activities towards various
triglycerides such as tributyrin (C4 : 0), tricaprylin (C8 : 0),
tricaprin (C10 : 0), trilaurin (C12 : 0), trimyristin (C14 : 0),
tripalmitin (C16 : 0) and tristearin (C18 : 0) at a final concentration
of 10mMand towards commercial oils (olive oil and
palm oil) were measured by the titrimetric method of Stuer
et al. (1986).
