Inhalation is the major route of exposure to toluene. However, it is also absorbed through dermal absorption and ingestion. About 60-75 of an absorbed dose in metabolized to benzolic acid, while 25-40% is excreted as unchanged toluene in expired air. The major metabolic pathway of toluene involves oxidation by cytochrome P-450, alcohol dehydrogenase and aldehyde dehydrogenase leading to benzoic acid which, upon conjugation with glycine,results in hippuric acid, the major urinary metabolite.
