of PLBs added per subculture period; in total, 659.5 PLBs formed after the third subculture; on average, 198.6 PLBs formed per subculture period; only 1.4% of PLBs browned. The second best culture system was when PLBs were cultured on semi-solid medium in Brand’s bot-tles (CS-1), while the poorest growth and proliferation of PLBs witha high incidence of PLB browning (as much as 2.8%) was observed inPLBs cultured in liquid medium in a 250-mL Erlenmeyer flask with 100 rpm shaking on a GFL 3017 orbital shaker (CS-4). Furthermore,half-strength MS medium containing 150 mL/L CW (M-1) was the most appropriate medium for growth and proliferation of D. ‘Zahra FR 62’ PLBs and gave a higher and more significant effect than other media: total FW of PLBs after the third subculture was 5.9 g; average FW of PLBs added per subculture period was 1.6 g; total number ofPLBs after the third subculture was 345.2; average number of PLBsper subculture period was 95.7; no PLBs browned (Table 2). VWmedium containing 0.05 mg/L BA (M-4) did not positively influence the growth and proliferation of PLBs and showed a high incidence of PLB browning (as high as 1.9%).
