2.1.2.2. Culture system set up. One, two or three-storey
hydroponic systems were set up by: (a) pouring 50 ml of
nutrient medium in the plastic box; (b) disposing the plastic ring
at the bottom of the box, the plastic sheet and the carrier
substrate (filter paper or cotton layer) were then placed,
respectively, on this ring; (c) placing 15 stem nodes on each storey; (d) pouring 2 ml of liquid medium onto the substrate to
provide the necessary nutrient for initial development of cutting
nodes.
The second and the third storey will be built by adding plastic
rings, plastic sheets, carrier substrates, explants and liquid
nutrient medium as described above [step (b–d)] if necessary.
The capillary robe was finally placed in the system from the top to
the bottom through a hole at the edge of each storey. To finish, the
cylindrical box were capped and covered with sterilized
polypropylene wrap to keep the system in the aseptic condition.
