Anthocyanin content
Anthocyanin content of the florets was determined using the
method of Nakamae and Nakamura (1983). Ten discs (0.53 cm in
diameter) from petals and sepals of first floret were bored using a
cork borer. The discs were washed in distilled water, placed in a 25
ml vial containing 10 ml methanol and 0.1% HCl (v/v) and then
stored at 25°C in darkness for 24 h. Absorbances of these solutions
were measured at 530 nm. Measurements were taken on day 1, 3
and 5.
