Bacterial strains screened positive for protease production in plate test were grown in liquid media (alkaline broth) for protease assay. A well grown bacterial suspension was centrifuged at 10,000×g for 10 min at 4 °C. The crude enzyme activity was assayed by the modified method of Tsuchida method [35] in cell free culture supernatant using 1% Casein as a substrate in 50 mM sodium phosphate buffer and Protein quantity was determined by the Lowry method [23]. In this process, Bovine serum albumin was used as standard. The crude enzyme activity was monitored at regular intervals (24 h) upto 7 days in triplicate times.
