Secretion of bacterial proteins is a necessary bottleneck to
overcome in production of recombinant proteins. In this review
we have outlined several strategies for enhancing
secretion, whether by genetic engineering or simply through
culture medium design. Culture medium could be designed to
allow for an increase in nonspecific secretion efficiency when
the mechanism is not entirely known. However, these strategies
may reduce cell viability, leading to an overall decrease in
protein secretion throughout the growth of the cell culture.
Other means to increase outer membrane permeability such as
deletion of Braun’s lipoprotein may be a step towards eliminating
the need for media additives, as the resulting “leaky
periplasm” phenotype allows increased protein secretion with
only a slight impact on growth
