2.11. Experimental design, data collection and statistical analyses
Thirty-nine plots (three replications for each treatment) were
set up using a randomized complete design. Each plot contained 21
single plants in 3 rows with 7 plants per row spaced 10 cm apart
with-in each row. Distance between rows was 40 cm, and between
plots 80 cm. Agricultural practices were carried out according to
the recommended production practices for the area. Plant height,
dry weight per plant (shoots were oven-dried for 2 days at 90 8C),
grain yield (seed weight) per plant, and 1000-seeds weight (14%
moisture content) were determined at the end of the experiment.
Plant height was measured from the soil surface to the apex, and
results were expressed in cm. Dry weight per plant were attained
by measuring of oven-dried (at 85 8C for 24 h) stem and leaves
samples, and results were expressed as g of dry weight per plant.
The variability of each analytical method was measured
repeating the analyses four times to confirm reproducibility.
Moreover, current results originate from representative experiments.
A quality control samples (blank-containing all reagents
except the test sample) was also analyzed together with the batch
of samples and recorded on a control chart. The results of all
control samples were within control limits thus confirming the
reliability of analysis.
An analysis of variance (ANOVA) was performed for the lentil
parameters (plant height, dry weight per plant, grain yield-seed
weight, and 1000-seeds weight) using the SPSS 11.0.1 for Windows
statistical package (SPSS, Chicago, USA). For comparison of the
means, the Fisher’s Protected LSD test at the 5% level was
employed. Data for TPC, TAC and specific phenolic compounds
were subjected to statistical analysis using Mann–Whitney U
nonparametric test (Statistica 6.0; StatSoft, Inc., USA).
