Total protein samples (200 mg) were loaded on 12% of SDSpolyacrylamide
gel electrophoresis, and transferred onto a 0.45 mm
nitrocellulose membrane (Bio-Rad, 162-0115). Membrane was
blocked at room temperature for 1 h with 5% non-fat milk in 1X
TBS-T, and then washed with 1% non-fat milk in 1X TBS-T. Membrane
was incubated at 4 °C for overnight with primary antibodies.
After thrice washing in 1% non-fat milk in 1X TBS-T for each
5 mins, membrane was incubated at room temperature for 1–2 h
with secondary horseradish peroxidase-conjugated antibodies
followed by washing with 1X TBS-T thrice for each 10 min. Protein
band was developed by Supersignal West Dura Extended Duration
Substrate kit (Thermo) and detected using a Fusion FX CCD camera
(Fisher Biotec). GAPDH was used to normalize protein loading.
Protein expression level was expressed as a relative ratio to
GPADH.
