In another experiment, zygotic embryos were cultured on MS
basal medium containing varying levels of Thidiazuron (TDZ). At
all the cytokinin levels tested, the embryos gave only a white
and spongy mass of non-embryogenic calluses after 2 weeks (data
not shown). The composition of the medium was found to be an
important factor for the induction of embryogenic callus. In our
study, the highest callus formation (93%) from the zygotic embryo
was recorded on half-strength MS medium supplemented with
1.1 mg l−1 of TDZ. However, the above growth regulator regime
resulted in only limited embryogenesis (data not shown). Though
the explants (zygotic embryo) treated with 2,4-D (2.0 mg l−1) produced
only 87% of callus, but the degree of embryogenesis was
higher (75.12%) compared with TDZ treatment, indicating that
callus induction media plays a vital role in determining somatic
embryogenesis in papaya (Table 1).
