Fresh samples, 0.5–1 kg each of the selected foods (exceptJerusalem artichoke) were randomly purchased from three retailedshops at Salaya market (as a representative market sold fresh foodsfrom Southern part of Thailand). A common variety of Jerusalemartichoke or Kaentawan was randomly collected from Khon KaenUniversity Agronomy Farm, Thailand. All food samples were keptin plastic bags and immediately transferred to the laboratory.Single composite samples of each food from three retailers wereprepared. The samples, except seeds and nuts, were washed withtap water until clean, blotted to remove excess water, weighed andthe inedible portion discarded (recorded % edible portion), rinsedwith deionised water once, blotted with clean tissue paper andthen air dried. The remaining edible portion was cut into small size( <1 cm on pieces) and homogenised by food processor (Tefal1Kaleo Blender, France). The homogenised samples were dividedinto 2 portions; one portion was determined as fresh sample formoisture analysis on the same day of sample preparation andanother portion was dried by a freeze-dryer (Lyovac1, Germany).The freeze-dried samples were homogenised by a grinder(Panasonic1MX-210GN, Japan) to obtain a fine powder. Theweight of sample before and after freeze drying was used forcalculation of inulin, FOS and sugar contents into fresh samplebasis. Seeds and nuts were ground by a Panasonic1grinder andanalysed for moisture. Each sample was kept in an acid-washedscrew cap polyethylene bottle until analysis. The single compositesamples of each food were analysed to identify potential sources ofinulin and fructo-oligosaccharides. Another two sets of thepotential food sources identified from the preliminary study weresampled again from two more markets in Bangkok, Thailand(Mahanak and Saphanmai markets, as a representative marketssold fresh foods from Central part and Northern part of Thailand,respectively). Two more varieties of Jerusalem artichoke werecollected again from the same places. They were processed andprepared as described in the preliminary study.
